I never said Charmander is bad but rather that Charmander is hard mode when it comes to Misty because Charmander/Charmeleon cannot resist her Water attacks(rather takes x2). That means you have to rely on the Pokémon you catch or utilize on your team to take her down. Pikachu is a solid option given it can use Thunder Wave to slow them down and has x2 Thunder Shock but is defensively frail and does not resist Water. Kanto is also set up to where your Grass type options are limited since the first one you encounter is Paras on Mount Moon, but Paras isn't exactly a powerhouse. It does know Stun Spore with a solid Physical attack stat but its slow as molasses and its Special could be better with no Grass type move to speak of for x2 damage. It does get Leech Life(20 power move) at 19 though, hence you could deal x2 Bug Damage to Starmie but that's all you really have. Your other Grass type option requires going north on Route 24/25 to go catch an Oddish(FR) or Bellsprout(LG) but requires battling your rival and Nugget Bridge Trainers before you can even get the opportunity to catch one. Training them is also a challenge because of the lack of opponents that Grass is strong against. Plus Oddish doesn't learn all its status moves until level 18 and doesn't get Petal Dance until level 39(was 33 in RBY). Bellsprout isn't much better since it has to go to level 19 to get Stun Spore but its Stun Spore + Wrap combination would normally be lethal in Gen 1 but Gen 3, Wrap is not the lock-down move it happens to be and hence is not the best option. Furthermore, you have to go to level 37 to get Razor Leaf(level 33 in RBY).
As for Nidoran, the version you have dictates which gender is more likely to spawn. Fire Red is more likely to get male while Leaf Green is more likely to get female. Yet if you really want to utilize the other gender, the game has an in-game trade where you trade the more common gender(in your case male Nidoran) for the less common one. The trainer is located in the Route 5 entrance to the Route 5-6 Underground passage where she will trade the less common Nidoran for the common one in your version. Meaning for Fire Red, she offers female Nidoran while in Leaf Green she offers male Nidoran. This helps players acquire the other Nidoran without having to spend time trying to hunt for them on Route 3 and 5. Furthermore, traded Pokemon grow faster so if you wanted to use Nidoqueen, this would be a pretty solid Trade but requires that you train it up. Also, Mount Moon has two Moon Stones, at least it does in Generation 1. If Generation 3 is anything like its previous iteration, then there's a hidden Moon Stone where you get the Fossils(I won't say explicitly where but its on that floor/area… if its there at all).
Another in-game Trade will be an Abra for a Mr.Mime(the only way to get it). So when you come across them, try to find and catch two for that in-game Trade. This one will be after going through Diglett Cave that is east of Vermilion. This cave will take you all the way back to Route 2 but in an area that is blocked off by a bush that you can Cut to return back to Pewter City if you need to grind up some Pokémon. In that area though is a House with the Trainer in question for Mr.Mime. You'll need Cut to head South so that you can acquire the Flash HM though. Flash is only needed for one area of the game and even then its optional but that area is Rock Tunnel on Route 10(Route 9 takes you there but you need Cut to access it, hence Cut is an essential HM). You have to go that way because you cannot go through Saffron to get to Celadon so you have to take the scenic route through the Tunnel to Lavender and from there head west to take another Underground passage that will get you to Celadon. Once in Celadon, you get the Fresh Water to unlock Saffron(permitting you to easily travel between Cerulean, Lavender, Saffron, Vermilion and Celadon). Upon reaching Celadon though, the Fly HM will nearly be in your grasp(I won't tell you where it is but only that its nearby).

The what now? All I mentioned is that Misty is a royal pain for Charmander users since their Starter is weak against her unless you're so overleveled that you can one-shot her Staryu with a strong Physical attack and manage to tank a Water Pulse(which I think is weaker than Bubblebeam in RBY so that's a slight nerf to her there) before dishing out retaliation since its doubtful you can outspeed her Starmie.
The rest was about the Nidoran lines and in-game trades. Both genders are available in either version but one gender is more common than the other one and the less common one can be obtained via an in-game trade. The other trade I talked about was for Mr.Mime, which you need to trade an Abra for(In Yellow they want a Clefairy). In-game trades are worthwhile since the traded Pokemon grow 50% faster.
Btw Izzy, you can find Poliwag/Poliwhirl once you get your hands on either the Good Rod or Super Rod. Since the Super Rod will come first if you head to Fuchsia via Route 12/13/14/15 or the "coastal" route, you can then go to pretty much any Fresh Water source(in any given town pretty much) and fish for them(Route 22 is a good place to get them). Once you get two Poliwhirl, you can then trade it for Jynx in Cerulean City.
